中性粒细胞是人类血液中的主要白细胞1,是先天免疫系统的关键参与者。它们大量到达有炎症或感染的组织2.在那里,中性粒细胞激活多种效应功能,例如吞噬作用3,4颗粒5,6和中性粒细胞细胞外陷阱(NET)的形成7。此外,中性粒细胞还参与适应性免疫反应8。中性粒细胞的经典观点认为它们是在骨髓中产生的具有预定反应的均质细胞9,10。然而,最近的研究表明,中性粒细胞是异质细胞,在健康和疾病状态下具有多种表型和功能状态 11,12,13,14,15。

在几个中性粒细胞亚群中,低密度中性粒细胞(LDN)因其内在特性以及它们在多种疾病中的数量增加而引起了人们的极大兴趣16,17,18。1986 年,在系统性红斑狼疮 (SLE) 患者的血液中发现了 LDN19。它们是在密度梯度20,21分离白细胞的过程之后检测到的。在密度培养基(例如 Ficoll-Paque)上离心血液或富含白细胞的血浆后,单核细胞和淋巴细胞(称为外周血单核细胞 [PBMC])在管的上部(低密度)部分形成一条带。中性粒细胞沉淀在管底部。在PBMC中,还发现了一些中性粒细胞;它们是LDN(图1)。健康个体的血液中存在少量 LDN22.然而,LDN 数量在多种免疫抑制和慢性炎症条件下显着增加,包括 SLE23,24、败血症25、牛皮癣26、哮喘27、幼年特发性关节炎28、抗中性粒细胞胞浆抗体 (ANCA) 相关血管炎29、HIV 感染30、疟疾31 和结核病32.尽管所有提到的病理中的 LDN 数量都有所增加,但 LDN 主要在 SLE17 的背景下进行研究。SLE 患者血液中的 LDN 似乎具有更强的形成 NET的能力 33、分泌大量促炎介质34 和激活 T 细胞24。然而,据报道,在其他情况下,例如癌症,LDN由成熟和未成熟的中性粒细胞35组成,具有T细胞抑制特性36,37,38。同样,在 COVID-19 患者中,据报道 LDN 也会抑制 T 细胞增殖39,但与 SLE 相反,LDN 似乎产生的 NET较少 39。因此,LDN的起源、组成和功能特性仍存在争议。

由于 LDN 与 PBMC 一起发现,因此研究它们在技术上很复杂。为了充分探索不同病理中的 LDN 特性,有必要对其进行纯化。LDN分离的常见程序是荧光细胞分选22,40,41,42,43,44,45。然而,细胞分选需要很长时间才能分离细胞。如果分选时间不够,这可能会导致细胞丢失或回收率低。此外,细胞承受过大的压力,在研究这些细胞的功能时导致不同的结果。分拣时间长也增加了实验程序的成本,需要专业人员。

本协议中的所有程序均遵循墨西哥国立自治大学 (UNAM) 生物医学研究所人类研究生物伦理委员会的指导方针。所有参与者都提供了知情同意书。

1. 从人血液中分离外周血单核细胞 (PBMC) 和中性粒细胞

  1. 通过静脉穿刺从健康的成年志愿者那里获取约 10 mL 血液。添加 10 U/mL 肝素作为抗凝剂。
    注意:将针头丢弃到生物危害针头处理容器中。注射器和其他与血液接触的材料应放入袋子中并在处理前进行高压灭菌。
  2. 在15mL锥形离心管中,在PBS中加入2mL的6%葡聚糖T500。然后通过将 10 mL 血液从管侧排出来添加。将管子倒置几次以混合血液和葡聚糖。
  3. 让试管静置 45 分钟,同时红细胞沉淀。富含白细胞的血浆出现在红细胞层上方。
  4. 在另一个 15 mL 离心管中,加入 5 mL 密度梯度培养基。小心地移液血浆,不要接触红细胞,并将其分层在培养基上。必须形成两个独立的阶段。
  5. 将试管在 4°C 下以 516 x g 离心 20 分钟。 单核细胞 (PBMC) 在血浆层和培养基层之间形成一条带。中性粒细胞在底部形成沉淀(图1)。
  6. PBMC的分离
    1. 通过抽吸消除PBMC顶部的血浆,而不接触细胞。从血浆和培养基之间的条带中收集细胞。确保吸出尽可能少的培养基。
    2. 将细胞放入 50 mL 锥形离心管中。加入 20 mL PBS。将试管在4°C下以400 x g 离心5分钟。
    3. 小心地吸出上清液并刮擦管子以分离细胞。加入 10 mL 冷 PBS 以重悬细胞。使用 Neubauer 室计算 PBMC。
      注意:请勿使用移液器重悬细胞,因为这可能会损坏细胞。
  7. 中性粒细胞的分离
    1. 去除密度梯度介质。通过刮擦管分离细胞并加入 10 mL 冷 PBS。
    2. 将细胞放入新鲜的 50 mL 离心管中,并在 4°C 下以 400 x g 离心 5 分钟。 吸出上清液并分离细胞,如步骤1.6.3中所述。
  8. 为了消除残留的红细胞,加入 10 mL 冷低渗溶液(0.2% NaCl,1% BSA,20 mM Hepes,pH = 7.4),并轻轻混合 1 分钟。
  9. 快速加入10mL冷高渗溶液(1.6%NaCl,1%BSA,20mM HEPES,pH = 7.4)使溶液等渗。
  10. 使用Neubauer室(纯度>95%)计数中性粒细胞。如步骤1.6.2中通过离心沉淀细胞。并将它们重悬于冷PBS中。将管子放在冰上。
    注意:据报道,中性粒细胞在体外的半衰期为8-12小时46,47,48。根据我们使用该方案的经验,中性粒细胞维持其功能约 6 至 8 小时。

2. 低密度中性粒细胞(LDN)的纯化

  1. 在4°C下以400 x g 离心PBMC5分钟。 除去上清液并将细胞重悬于 120 μL 冷洗缓冲液(PBS 中的 1% BSA)中。
  2. 加入 35 μL CD66b 磁性微珠。将混合物在4°C的黑暗中孵育30分钟。每 10 分钟轻轻混合一次,以防止细胞聚集。
  3. 加入 1 mL 冷洗缓冲液。将细胞放入微量离心机中,以 400 x g 旋转 3 分钟。
  4. 除去上清液,通过刮管分离细胞沉淀,并将细胞重悬于 1 mL 洗涤缓冲液中。
  5. 将磁分离柱放在磁铁上。向色谱柱中加入 0.5 mL 洗涤缓冲液,让它通过色谱柱。
  6. 将细胞 (1 mL) 转移到色谱柱上。让缓冲液一滴一滴地穿过色谱柱。
  7. 将 0.5 mL 洗涤缓冲液加入色谱柱中,让其通过。将另外 0.5 mL 洗涤缓冲液加入色谱柱中。
  8. 从磁铁上取下色谱柱并将其放入微量离心管中。向色谱柱中加入 1 mL 洗涤缓冲液。
  9. 将柱塞插入色谱柱顶部,轻轻施加压力以洗脱细胞。取下柱塞,将色谱柱放在新的微量离心管顶部。
  10. 再加入 1 mL 洗涤缓冲液。将柱塞插入色谱柱顶部,轻轻施加压力以洗脱细胞。
  11. 将两个试管放入微量离心机中,并以 800 x g 旋转 3 分钟。最后,将两个管中的细胞(LDN)重悬于1mL冷PBS中。保持冰上。
    注意:前 2 mL 流通包含其余的 PBMC。这些细胞可用于其他研究。

3. 多色流式细胞术

  1. 将纯化的细胞以 1 x 106 细胞/mL 重悬于标记缓冲液(PBS 中的 1% FBS)中。将 250 μL 细胞加入 1.5 mL 微量离心管中。
  2. 添加相应的中性粒细胞膜分子抗体(有关详细信息,请参阅 材料表)。将细胞在4°C下孵育30分钟,保护它们免受光照。
  3. 加入 1 mL PBS。将试管放入微量离心机中,以 800 x g 旋转 3 分钟。

4. 活性氧(ROS)的检测

  1. 在 1.5 mL 微量离心管中,加入 2.5 x 105 个细胞。将试管放入微量离心机中,以800× g 旋转3分钟。
  2. 吸出上清液,刮管分离细胞,并将细胞重悬于PBS中的100μL 15μM二氢罗丹明123中。
  3. 将避光细胞在37°C下孵育20分钟。加入 1 mL PBS。
  4. 将试管放入微量离心机中,以800× g 旋转3分钟。吸出上清液,刮管以分离细胞,并将它们重悬于 PBS 中的 100 μL 50 nM 佛波醇 12-肉豆蔻酸酯 13-乙酸酯 (PMA) 中。
  5. 将避光细胞在37°C下孵育45分钟。加入 1 mL PBS。
  6. 将试管放入微量离心机中,以 800 x g 旋转 3 分钟。吸出上清液,刮管分离细胞,并将它们重悬于 PBS 中的 0.5 mL 1% 多聚甲醛中。

5. 中性粒细胞细胞外陷阱 (NET) 的可视化

  1. 将盖玻片放入 12 孔板的孔中,并用 10 μg/mL 聚-L-赖氨酸覆盖。将板在4°C下轻轻搅拌孵育过夜。
  2. 除去聚-L-赖氨酸,并用PBS洗涤盖玻片,在室温下轻轻搅拌将板孵育5分钟。用 PBS 再清洗盖玻片两次。
  3. 取出PBS,将板放入层流罩内2小时,让盖玻片干燥。
  4. 将 LDN 以 1 x 106 细胞/mL 重悬于含有 5% FBS 的 RPMI-1640 培养基中。取 350 μL(3.5 x 105 个细胞)细胞悬液,并将它们放入含有盖玻片的 12 孔板的孔中。
  5. 将板在37°C的5%CO2 培养箱中保持30分钟。 向每个孔中加入 3.5 μL 用 PBS 稀释的 5 μM PMA(PMA 的终浓度为 50 nM)。
  6. 将板在37°C的5%CO2 培养箱中保持4小时。 在PBS中加入350μL8%多聚甲醛,并将板在37°C的5%CO2 培养箱中过夜。
  7. 如前所述49,在试管架上放置透明薄膜,以便在管孔上形成孔。
  8. 用各种溶液填充每个孔,以清洗或染色盖玻片,形成大液滴。取下盖玻片,将它们倒置在一滴水上 5 分钟。以同样的方式,用水再清洗盖玻片三次。
  9. 将盖玻片倒置在一滴封闭缓冲液(PBS 中的 5% BSA)上,并在室温下孵育 20 分钟。
  10. 将盖玻片转移到封闭缓冲液中的一滴相应一抗(例如,抗弹性蛋白酶或抗瓜氨酸)中,并在室温下孵育 60 分钟。
  11. 在 PBS 中的 0.01% 吐温-20 中洗涤盖玻片 2 次。将盖玻片转移到含有 150 nM DAPI 的封闭缓冲液中的一滴相应二抗中,并在室温下避光孵育 60 分钟。
  12. 在 PBS 中的 0.01% 吐温-20 中洗涤盖玻片 2 次。将一滴抗淬灭封片介质滴在载玻片上,然后将盖玻片倒置。
  13. 用指甲油密封周边的盖玻片。将安装的载玻片储存在 4 °C 下,避光。使用荧光显微镜可视化 NET。

6. 检测中性粒细胞细胞外陷阱 (NET)

  1. 将细胞以 5 x 105 细胞/mL 重悬于 500 nM Sytox Green 中,在用 5% FBS 稀释的 RPMI-1640 培养基中。
  2. 取 100 μL(5 x 104 个细胞)细胞悬液,并将它们放入 96 孔组织培养板的孔中。
  3. 将板在37°C的5%CO2 培养箱中保持20分钟。 向每个孔中加入20μL在RPMI-1640培养基中稀释的300nM PMA(PMA的终浓度为50nM)。
  4. 将板转移到预热的酶标仪上。将板在35°C下孵育4小时,每5分钟从板底部进行荧光测量(使用485nm激发和528nm发射滤光片)。
