肠道上皮是一种动态组织,持续自我更新和分化,以维持肠道稳态并促进黏膜损伤后的再生 1,2。肠道上皮细胞起源于肠道干细胞(ISCs),它们与多种微环境因子345678相互作用。上皮下区室,主要是固有层和黏膜肌,由不同谱系的多样细胞群组成9.在这些细胞群体中,肠道间充质间质细胞(iMSCs),包括成纤维细胞和肌纤维母细胞,产生营养和生长因子,调节ISC干的生长、增殖和分化,以调节上皮稳态和再生6,7,10,11。

为了研究这些细胞相互作用,近期研究开发了新方法,揭示驱动肠道上皮细胞与iMSC交互作用的可能机制。将类器官与基质细胞或免疫细胞结合的共培养系统,推动了我们对上皮-上皮下细胞在发育、再生、药物筛选及疾病建模过程中相互作用的理解,12,13,14,15,16,17。然而,大多数类器官共培养研究使用了小鼠组织,这些组织往往无法有效模拟人体生理和疾病表型,限制了其临床转化相关性18

在大多数小鼠肠道分离方案中,上皮细胞和基质细胞分别取自不同的组织片段,这可能产生反映略微特征的样本,可能影响实验间的重复性19,20。目前,尚无统一优化的方案用于从同一组织样本中分离猪肠道窝和iMSCs。此外,获取合适人肠样本的实际局限性使许多研究者依赖不同器官(如具有皮肤成细胞的人类肠)不匹配的细胞类型来评估上皮-间充质细胞交叉,这可能干扰数据解释(21,22,23)。

尽管最近发表的一项人类肠道活检方案通过残留上皮枯竭组织的生长分离出了窝穴并获得成纤维细胞,但猪中缺乏类似的标准化方法24。由于猪在解剖和生理学上与人类相似,被用作转化研究的合适动物模型 2,18,25,26,27,28,29。因此,该方法采用了一种顺序分离协议,基于肠道的生理、解剖和形态结构,从同一猪空肠节段回收肠道隐窝和iMSCs该程序为分离未来猪肠道共培养研究的隐穴和iMSCs提供了可重复的框架。

从马里兰大学学院公园分校猪舍中切除的21天大幼猪近端空肠中解剖了一段5厘米的片段。用于宰牲幼猪的程序(R-6月22日)遵循了大学机构动物护理与使用委员会批准的动物护理和使用政策指南。在进行分离步骤之前,协议概要见 图1,用于顺序分离猪肠道隐穴和iMSCs的试剂和设备列表见 材料表

1. 隔离前准备阶段

  1. 准备每5厘米组织段30毫升洗涤液,置于50毫升锥形管中。
  2. 根据采集的组织样本数量调整体积。
    注意:请查看 表1 上的洗涤缓冲剂成分。所有组织制备和细胞分离步骤均在工作台上完成。

2. 肠道组织准备阶段

  1. 将切除的肠组织放入30毫升的洗涤液中,置于50毫升锥形管中。
    注意:切除的组织可在使用前放入冷水冲洗缓冲液中一小时。
  2. 用21克注射器用冷却洗涤液冲洗3×小时,去除腔内成分。
  3. 将组织放置在消毒过的聚苯乙烯泡沫表面上,小心地将肠系膜和脂肪组织从浆膜中取出。
  4. 用手术刀纵向解剖组织,打开腔内(图2A)。
  5. 将部分清理后的解剖组织转移到新鲜的洗涤液中,洗涤两次,或直到缓冲液变透明。
  6. 将组织放入100毫米培养皿的洗涤缓冲液中,用新的手术刀片切成10-15毫米的片(图2B,C)。这一步增加了暴露于消化缓冲液的表面积。
  7. 将切碎的组织转移到新的50毫升锥形管中。
  8. 用洗涤缓冲液轻轻清洗,让碎碎组织通过重力沉降约30秒(见图2D)。
  9. 轻轻吸取上清液,只留足够的缓冲液覆盖锥形管内的组织碎片。
  10. 上皮解离阶段
    1. 加入30毫升预热上皮解离缓冲液(见表2),在37°C的静态珠浴或约9× 的摇晃水浴中孵育20-30分钟。
    2. 每15分钟轻轻摇晃管子,以增加窝穴从上皮膜的释放。
    3. 让组织沉降,并用血清学移液器轻轻吸取上清液,只留足够的缓冲液覆盖锥形管内的沉积组织。
    4. 向沉积组织加入30毫升新鲜洗涤液,用力摇晃以释放绒毛、窝穴和杂质(见图3A)。让组织残留物沉淀,然后轻柔地将含有隐穴段的洗涤缓冲液上清液通过100微米的细胞滤网,倒入一个新的50毫升锥形管中。
      1. 重复添加新鲜洗涤缓冲液到残留组织中并倒出三次(3×)。将含有隐穴的10微升洗涤缓冲液移入100毫米培养皿,在立体显微镜下检查隐窝密度。
        注意:每次洗涤后,每个洗刷缓冲区都包含地穴。
    5. 第三次洗涤后,包含墓穴的洗涤缓冲区应保持清澈,杂质极少(见图3B)。
      注意:将带有细胞外基质(ECM)的组织残余保存在锥形管内,以便后续的iMSC分离处理。
    6. 在室温下以150× 离心分离地穴悬浮液2分钟,以沉淀地窖碎片。
    7. 抽吸并重新悬浮于上皮培养基中。
    8. 再次以150× 离心分离,收集墓穴碎片颗粒。
    9. 加入上皮培养基以重新悬浮巢穴,并用1000微升移液器尖端移液5×。
    10. 将约100个重新悬浮的隐穴种子到预先制备的胶原蛋白1型水凝胶涂层6孔板上,培养并扩展为二维(2D)肠状单层。
      注意:请查看 表3中的胶原水凝胶涂层平板制备。最终胶原蛋白1型浓度应为1.0 mg/mL。避免对过密的墓穴或碎屑进行镀层,以防止酵母菌或真菌污染。在洗涤液和培养基中加入抗生素-抗真菌(anti-anti)应提供保护。
    11. 在37°C加湿的5%二氧化碳培养箱中培养前,先在倒相对比显微镜下观察细胞。
    12. 播种后12小时内更换培养基,以去除漂浮的杂质并减少细菌污染。
    13. 使用上皮培养基(表4)培养肠类菌株,并每隔一天更换培养基,直到约第8天,扩展后的二维单层达到≥85%汇合度进行亚培养。
  11. 细胞外基质解离阶段
    注意:继续分离过程,使用含有细胞外基质的剩余组织片段(见图3C图4A)。
    注意:虽然上皮解离缓冲液中的EDTA可以抑制胶原酶活性,但前三个洗涤步骤已去除所有EDTA和DTT。因此,胶原酶活性在后续过程中不应受到影响。
    1. 将剩余组织再次切碎为小块,然后分离本有层的iMSCs。
    2. 向组织中加入25毫升细胞外消化缓冲液(见表5),并在静态珠浴中37°C下孵育15-20分钟(见图4B)。
    3. 孵化后用力摇晃。
      注意:上清液会变得浑浊,漂浮组织碎片减少。在倒置显微镜下移液10微升,检查上清液中iMSC的密度。如果观察到的iMSC密度较低,继续在珠浴中孵育15分钟。
    4. 在步骤2.2.3剧烈摇晃后,轻轻将上清液通过100微米的细胞筛过滤到新的锥形管中,以减少杂质。
    5. 在室温下以280× 离心分离细胞悬浮液5分钟,沉淀iMSCs。
    6. 抽取上清液,并将iMSCs重新悬浮到间充质培养基中(见 表6中的iMSC培养基)。
    7. 再次以280× 离心分离分离,去除解离缓冲液。
    8. 在间充质培养基中抽吸并重新悬浮iMSCs。
    9. 将iMSCs置于T75培养瓶中,每个瓶约5个5个细胞×10个5 细胞。
      注意:确保镀层细胞覆盖烧瓶表面面积约60%,因为间充质细胞需要中等至较高的播种密度以实现最佳生长。
    10. 在电镀后12小时内更换培养基,以去除多余的杂质并防止细菌污染。
    11. 在播种后第2天,用倒置相对比显微镜观察类成纤维细胞。
      注意:间充质培养基应隔天更换一次。iMSC应达到约85%的汇合度,并在第4天准备进行亚培养。

3. 免疫细胞化学测定

  1. 在96孔板中用间充质培养基培养约5,000个iMSCs,培养2-3天,直到细胞达到约80%的汇合度。
  2. 在室温31下,轻轻抽吸培养基,并用100微升1×PBS每孔冲洗两次培养的iMSCs。所有关于免疫染色试剂盒的相关资料均见 材料表
  3. 每孔加入100微升4%对甲醛(PFA)固定培养的iMSCs,并在室温静态台面上培养30分钟。
  4. 用100微升0.1% Tween-20/PBS(PBST)洗涤固定培养的iMSC两次,每次5分钟,然后用100微升0.5% Triton X-100/PBS渗透30分钟。
  5. 每孔添加100微升5%牛血清白蛋白(BSA)/PBST,以阻断固定细胞上的非特异性结合。
    注意:在室温下放置1小时后,用100微升PBST洗涤两周,每次5分钟。
  6. 每个孔槽加入100微升稀释的一级抗体(1:250;即1微升一级抗体溶于250微升抗体稀释剂中),并在4°C下培养细胞过夜。 主要抗体为PDGFRα和CD81。
    注意:仅添加抗体稀释剂,不添加原抗,包含阴性对照井。
  7. 第二天,用100微升PBST冲洗一次原抗,每次5分钟。
  8. 将二级抗体稀释于1×PBS,使用以下比例:Alexa Fluor Plus 555(1 μL:100 μL)、Alexa Fluor Plus 647(1 μL:100 μL)和DAPI(1 μL:800 μL)。
  9. 用稀释的二级抗体在室温下对细胞进行反染1小时。
  10. 用1×PBS清洗细胞两遍,每次5分钟。
  11. 每孔加入100微升装载介质,覆盖板材,用铝箔包裹,在4°C下保存长达6个月,直到荧光成像。

4. RNA分离与qPCR测定

  1. 利用酸性硫氰酸镍-酚-氯仿分离技术,从T75瓶中培养的iMSCs中提取总mRNA。
    注意:使用分光光度计测定RNA浓度和纯度。
  2. 将总mRNA逆转录为cDNA,作为定量逆转录聚合酶链反应(qRT-PCR)测试的模板,以验证选定的Sus scrofa基因引物GAPDH(前向:ATCCTGGGTACACTGAGGAC;反向:AAGTGGTCGTTGAGGGCAATG),如先前研究所述2,28

在上皮解离过程中,乙二胺四醋酸(EDTA)螯合钙离子以破坏钙依赖性细胞间粘连,而二硫糖醇(DTT)则切断黏蛋白糖蛋白中的二硫键以促进黏液溶解。因此,螯合化合物(EDTA)和还原剂(DTT)的结合使用,使得隐穴能够有效释放上皮膜,同时不破坏细胞外基质(ECM)结构 2,32见图3)。隐室成功从上皮层释放,ECM干扰极小(见图3B,C)。在肠上皮培养基中培养时,分离的窝体附着并扩展为胶原蛋白包覆水凝胶表面的肠状单层,培养基每隔一天更换一次(见图5A)。胶原水凝胶为肠道小肠单层提供必要的结构支持,重现体内结构组织,并模拟上皮生长所需的结构。
