3d Refinement

3D refinement is a computational procedure that improves a three-dimensional model of a biomolecule by optimizing its agreement with experimental structural data. In biochemistry, it commonly uses iterative image alignment and reconstruction: particle orientations, translations, and map or model parameters are adjusted and compared with observed data until the refinement converges on a more consistent structure. This process can sharpen density, resolve molecular features, and support interpretation of protein complexes, nucleic acids, and other macromolecular assemblies. Reliable 3D refinement helps researchers evaluate conformational states, identify ligand-binding environments, and build structural models that guide mechanistic studies and experimental design.

Here, we present a protocol to isolate microglia from postnatal mouse pups (day 1) for in vitro experimentation. This improvised method of isolation generates both high yield and purity, a significant advantage over alternate methods that allows broad range experimentation for the purposes of elucidating microglial biology.

2024

Training protocols involving positive reinforcement for routine husbandry procedures in rabbits were developed and proved to be successful, as the rabbits reliably showed the trained behaviors at the weekly general examination. Picking the rabbits up with the transport box was less aversive than the conventional handling technique.
