Bap1-deubad

BAP1-DEUBAD is a protein interaction module that regulates the activity of the BAP1 deubiquitinase, an enzyme involved in removing ubiquitin from target proteins and controlling chromatin function. The DEUBAD region of ASXL proteins binds BAP1, stabilizes its catalytic domain, and promotes deubiquitination of substrates such as monoubiquitinated histone H2A. This interaction is central to the BAP1-Polycomb repressive deubiquitinase complex, which influences gene regulation, cell identity, and genome maintenance. Studying BAP1-DEUBAD structure and function helps explain how mutations in BAP1 or ASXL proteins disrupt ubiquitin signaling and contribute to developmental disorders and cancer.
