Leukocyte Elastase Inhibitor

Leukocyte elastase inhibitor is an endogenous protein that regulates leukocyte elastase, a serine protease released primarily by neutrophils during inflammatory responses. It works by binding the enzyme’s catalytic region and blocking its access to protein substrates, limiting the breakdown of elastin and other extracellular-matrix components while helping protect surrounding tissue from excessive proteolysis. In biochemistry, studying this inhibitor clarifies how protease activity is balanced during inflammation and tissue repair. Its interactions with leukocyte elastase support research into pulmonary and inflammatory disorders, provide tools for measuring protease regulation, and inform the development of selective inhibitors or related therapeutic strategies.

This video shows how to induce abdominal aortic aneurysms (AAA) in mice via transient intraluminal infusion of porcine pancreatic elastase into the infrarenal segment of the abdominal aorta. The model has the ability to add broad insight into the pathobiology of AAA due to the emergence of numerous transgenic and gene knockout mice.

A rapid method to obtain infiltrating leukocytes from the murine brain is described. This method utilizes a continuous Percoll gradient and discontinuous Ficoll gradient to select and purify the leukocyte-enriched layer. Isolated leukocytes may then be characterized by flow cytometric measurements.

After excising a segment of the right CCA, the pouch is treated with elastase, then sutured to the left CCA to restore blood flow. Marginating-hepatic leukocytes exhibit unique characteristics and distinct immunological functions compared to other leukocyte populations. Here we describe a method for selective harvesting of this specific hepatic cell population, through forced perfusion of the liver of rats or mice. Marginating-hepatic leukocytes seem critical in determining susceptibility to hepatic-related diseases and metastases.
