Nutrient Depletion

Nutrient depletion is the progressive reduction of essential nutrients in a biological system, medium, tissue, or environment, potentially limiting growth, metabolism, and cellular function. It occurs when nutrient uptake and consumption exceed replenishment, lowering the availability of substrates such as carbon sources, nitrogen, minerals, or metabolic cofactors and altering enzyme activity and metabolic flux. In biochemistry, monitoring nutrient depletion helps researchers interpret changes in cell growth, energy production, biosynthesis, and stress responses. The concept is important in cell culture, microbial fermentation, nutrition research, and ecosystem studies, where nutrient limitation can shape experimental outcomes and guide media optimization or metabolic engineering.

Here, we present an easy-to-follow protocol to establish a successful hydroponic system for plant nutrition studies. This protocol has been extensively tested in Arabidopsis and can easily be adapted to other plant species to study specific nutritional requirements or the effect of non-essential elements on plant growth and development.

Macrophages play a central role in homeostasis and pathology in many tissues. The protocol presented here describes methods for depleting macrophages in vivo, deriving polarized macrophages from bone marrow aspirates, and adoptively transferring macrophages into mice. These techniques allow determination of the role that polarized macrophages play in health and disease.
