Peptide Functionalization

Peptide functionalization is the deliberate chemical modification of a peptide’s termini or amino acid side chains to introduce new properties, making it valuable for biochemical research and therapeutic design. It typically relies on selective coupling reactions, in which reactive groups such as amines, thiols, or carboxylates are linked to labels, lipids, polymers, or other molecular components under controlled conditions, often with protecting groups to limit unwanted reactions. Functionalized peptides can support fluorescence labeling, immobilization on surfaces, drug delivery, and improved stability or activity. These modifications help researchers investigate peptide structure and interactions while enabling the development of biosensors, diagnostics, and peptide-based therapeutics.

Techniques to dissect the mechanisms underlying the secretion of HIV-1 Nef in exosomes are described. Specific short peptides derived from Nef and protein transfection were exploited to determine the structure, function, and binding partners of Nef’s Secretion Modification Region. These procedures have general relevance in many mechanistic studies.
