3d Printed Molds

3D printed molds are digitally designed forms fabricated layer by layer to shape materials into precise, repeatable structures, making them valuable for rapid prototyping and customized manufacturing. In bioengineering, a computer-aided model is converted into printing instructions, and deposited or cured material builds the mold geometry; selected materials and surface features then control the casting, molding, or gel formation process. These molds can produce tissue-engineering scaffolds, microfluidic devices, cell culture platforms, and patient-specific biomedical components. Their accessible design-to-fabrication workflow supports complex architectures, shortens development cycles, and enables researchers to test biological systems with greater geometric control.

A bioprinter was used to create patterned hydrogels based on a sacrificial mold. The poloxamer mold was backfilled with a second hydrogel and then eluted, leaving voids which were filled with a third hydrogel. This method uses fast elution and good printability of poloxamer to generate complex architectures from biopolymers.

This protocol describes a platform for fabricating self-assembled tissue rings in variable sizes using a customized 3D-printed plastic mold. PDMS negatives are cured in the 3D-printed mold; then agarose is cast in the cured PDMS negatives. Cells are seeded into the resulting agarose wells where they aggregate into tissue rings.
