Cell Morphology Control

Cell morphology control is the deliberate regulation of a cell’s shape, size, and structural organization to influence its behavior in engineered biological systems. Cells sense physical and biochemical cues, including substrate stiffness, surface adhesion, and geometric confinement, which alter cytoskeletal tension, focal adhesion formation, and intracellular signaling. Bioengineers use micropatterned substrates, tunable biomaterials, controlled cell density, and fluidic forces to guide spreading, elongation, alignment, or three-dimensional organization. Controlling morphology helps link cell structure to functions such as migration, differentiation, proliferation, and tissue formation, supporting the design of disease models, biomaterials, organ-on-chip platforms, and regenerative medicine strategies.

We present a protocol that permits to view and to quantitatively asses the morphology of the dendritic tree of individual Purkinje cells grown in organotypic cerebellar slice cultures. This protocol is intended to promote studies on the mechanisms of Purkinje cell dendritic development.

Cortical networks are controlled by a small, but diverse set of inhibitory interneurons. Functional investigation of interneurons therefore requires targeted recording and rigorous identification. Described here is a combined approach involving whole-cell recordings from single or synaptically-coupled pairs of neurons with intracellular labeling, post-hoc morphological and immunocytochemical analysis.
