Cryopreservation Solution

A cryopreservation solution is a specially formulated medium that protects cells, tissues, or biological materials during freezing and thawing, supporting their long-term storage for bioengineering and biomedical research. It typically contains cryoprotective agents that reduce ice crystal formation, limit cellular dehydration and osmotic stress, and help preserve membrane and protein integrity as temperature decreases. Used with controlled cooling and storage at very low temperatures, these solutions help maintain viability and function after thawing. Their performance influences the success of cell banking, tissue engineering, regenerative medicine, organoid research, and the development of engineered biological products.

The isolation of neonatal rat cardiomyocytes is a time consuming and unpredictable procedure. This study describes methods for cryopreservation and thawing of neonatal rat cardiomyocytes that allows for more efficient use of cells. The thawed NRCMs can be used for various experiments without the need for performing isolations each time.

Here we demonstrate the newly developed I•Cryo kit for mouse sperm cryopreservation. Two-cell stage embryo development with frozen-thawed sperm was improved consistently in 5 mouse strains with the use of this kit. Over a 1.5 year period, 49 genetically modified mouse lines were archived by sperm cryopreservation with the I•Cryo kit and later successfully recovered by IVF.

Preimplantation embryos may be cryopreserved after placement into a hypertonic cryoprotective solution to cause cellular dehydration. After equilibration, ice crystal formation is induced in the solution surrounding the embryo. Further dehydration occurs as the embryo is slowly cooled to subzero temperatures before plunging into liquid nitrogen for storage.
