Diffuse Reflectance Spectroscopy

Diffuse reflectance spectroscopy is an optical measurement technique that characterizes materials by analyzing light scattered and reflected from their surfaces or near-surface regions. It illuminates a sample with broadband or selected-wavelength light, while a detector collects diffusely reflected photons whose wavelength-dependent intensity changes with absorption and scattering by the sample’s chemical and structural components. In bioengineering, this approach supports noninvasive tissue characterization, monitoring of biological processes, and assessment of biomaterials and engineered constructs. Because measurements can be rapid and require minimal sample preparation, diffuse reflectance spectroscopy contributes to biomedical sensing, quality control, and the development of optical tools for research and clinical applications.

This protocol describes how the use of diffuse polarization spectroscopy can improve the clinical usefulness of the capillary refill test. We suggest a more detailed analysis of the course of the capillary refill in healthy volunteers using diffuse reflectance spectroscopy videos and new informatic endpoints.

Formulation of stable, functional inks is critical to expanding the applications of additive manufacturing. In turn, knowledge of the mechanisms of dispersant/particle bonding is required for effective ink formulation. Diffuse Reflectance Fourier Transform Infrared Spectroscopy (DRIFTS) is presented as a simple, inexpensive way to gain insight into these mechanisms.

We present a diffuse optical spectroscopic (DOS) approach that provides quantitative optical biomarkers of skin response to radiation. We describe DOS instrumentation design, optical parameters extraction algorithms and the animal handling procedures required to yield representative data from a pre-clinical mouse model of radiation induced erythema.
