Lamp Primer Sets

LAMP primer sets are coordinated oligonucleotide designs used in loop-mediated isothermal amplification, a rapid nucleic acid amplification method that enables sensitive detection without thermal cycling. Typically, four core primers recognize multiple regions of a target sequence, while loop primers can accelerate amplification; a strand-displacing DNA polymerase then generates loop-containing products at a constant temperature. In bioengineering, careful primer design supports pathogen detection, genetic identification, environmental monitoring, and portable biosensor development. Their high target specificity and compatibility with simple heating equipment make LAMP primer sets valuable for translating molecular assays from centralized laboratories to field and point-of-care settings.

Diagnosis of Eimeria infection in chickens remains demanding. Parasite morphology- and host pathology-led approaches are commonly inconclusive, while molecular approaches based on PCR have proven demanding in cost and expertise. The aim of this protocol is to establish loop-mediated isothermal amplification (LAMP) as a straightforward molecular diagnostic for eimerian infection.

Primer-Free Aptamer Selection Using A Random DNA Library

SELEX protocols comprise multiple rounds of selection, each of which require regeneration of bound ligands, which in turn require fixed primer sequences flanking the random library regions. These fixed primer sequences can interfere with the selection process (false positives and negatives). Here we present a primer-free protocol.

Primer Extension Capture: Targeted Sequence Retrieval from Heavily Degraded DNA Sources

We present a method of targeted ancient DNA sequence retrieval, which we used to reconstruct the complete mitochondrial genomes of five Neandertal individuals. Comparison of these sequences with present day humans suggests that Neandertals had a long term low effective population size.

Laboratory Protocol for Genetic Gut Content Analyses of Aquatic Macroinvertebrates Using Group-specific rDNA Primers

Most common gut content analyses of macroinvertebrates are visual. Requiring intense knowledge about morphological diversity of prey organisms, they miss soft bodied prey and, due to strong comminution of prey, are nearly impossible for some organisms, including amphipods. We provide detailed, novel genetic approaches for macroinvertebrate prey identification in the diet of amphipods.
