Nhs Ester Chemistry

NHS ester chemistry uses N-hydroxysuccinimide esters as activated carboxylic acid derivatives to form stable covalent bonds with primary amines, making it important for biomolecule modification and bioengineering. In mildly basic aqueous buffers, an amine nucleophile attacks the activated carbonyl, displacing N-hydroxysuccinimide and producing an amide bond, while competing hydrolysis can reduce labeling efficiency. This reaction supports protein conjugation, fluorescent labeling, crosslinking, surface functionalization, and attachment of biomolecules to polymeric or hydrogel materials. Controlling pH, reagent ratios, reaction time, and water exposure helps improve coupling yield and preserve biological function.
