Seed Sterilization Method

Seed sterilization is a surface decontamination method that reduces bacteria, fungi, and other microorganisms on seed coats before laboratory culture or bioengineering experiments. The process typically combines washing with a surfactant, brief exposure to ethanol, and treatment with an oxidizing disinfectant such as sodium hypochlorite, followed by sterile-water rinses to remove residual chemicals. By limiting microbial growth without substantially impairing seed viability, sterilization supports aseptic germination, plant tissue culture, genetic transformation, and propagation of engineered lines. Consistent sterilization improves culture reliability, reduces contamination-related loss, and helps researchers distinguish biological responses from effects caused by unwanted microorganisms.

The objective of this study was to determine the effects of bleach and chlorine gas sterilization on seed germination of a range of Arabidopsis genotypes grown on sterile media. Optimized sterilization protocols have been developed to prevent the growth of microbial contaminants while providing satisfactory seed survival.

We synthesized star shaped gold nanostars using a silver seed mediated growth method. The diameter of the nanostars ranges from 200 to 300 nm and the number of tips vary from 7 to 10. The nanoparticles have a broad surface plasmon resonance mode centered in the near infrared.
