Spent Media Analysis

Spent media analysis is the examination of culture medium after cells or microorganisms have grown in it, providing a noninvasive view of cellular activity and bioprocess performance. By measuring changes in nutrients, metabolites, dissolved gases, and secreted products, researchers can infer consumption rates, metabolic pathways, growth conditions, and cellular stress. In bioengineering, this approach supports optimization of cell culture and bioreactor processes, helps identify limiting substrates or toxic byproducts, and informs media formulation and process control. Tracking these chemical signatures can improve productivity, reproducibility, and scale-up while reducing the need to disrupt or harvest the cells being studied.

The present study reports a protocol for chromosome screening of human embryos that uses spent culture medium, which avoids embryo biopsy and enables chromosome ploidy identification using NGS. The present article presents the detailed procedure, including the preparation of culture medium, whole genome amplification (WGA), next-generation sequencing (NGS) library preparation, and data analysis.
