Absorbance Spectra

Absorbance spectra describe how strongly a substance absorbs light across different wavelengths, providing a characteristic optical profile for identifying and quantifying chemical or biological samples. In a spectrophotometer, light passes through the sample, and molecules absorb specific wavelengths when photon energy matches electronic transitions; the measured absorbance can be related to concentration through the Beer–Lambert law. In biological techniques, absorbance spectra help measure nucleic acids and proteins, characterize pigments and other biomolecules, assess sample purity, and monitor enzyme or cell-based reactions. These measurements support routine laboratory analysis and quantitative research in biochemistry, molecular biology, and biotechnology.

Source: Michael G. Benton and Kerry M. Dooley, Department of Chemical Engineering, Louisiana State University, Baton Rouge, LA Gas absorbers are used to remove contaminants from gas streams. Multiple designs are used to accomplish this objective1. A packed bed column uses gas and liquid streams running counter to each other in a column packed with loose packing materials, such as ceramics, metals, and plastics, or structured packing1. The packed bed uses surface area created by the packing to...

Emission Spectra

When solids, liquids, or condensed gases are heated sufficiently, they radiate some of the excess energy as light. Photons produced in this manner have a range of energies, and thereby produce a continuous spectrum in which an unbroken series of wavelengths is present. In contrast to continuous spectra, light can also occur as discrete or line spectra having very narrow linewidths interspersed throughout the spectral regions. Exciting a gas at low partial pressure using an electrical current,...
