Annealing Temperature Optimization

Annealing temperature optimization is the process of selecting the temperature that promotes efficient, specific binding of primers to a target DNA sequence during polymerase chain reaction (PCR). The temperature influences primer-template hybridization: conditions that are too low can permit nonspecific binding, whereas conditions that are too high may reduce primer attachment and product yield. Researchers commonly evaluate a temperature range using gradient PCR and assess amplification by product presence, size, and specificity. Optimized annealing conditions improve the reliability of DNA amplification for cloning, genotyping, sequencing, diagnostics, and other biological techniques.

This video demonstrates the gradient polymerase chain reaction for optimization of the annealing temperature of primers, which sets gradient annealing temperature along wells. The effect of different annealing temperatures is determined by the yield of amplified products that helps to optimize the reaction condition.

Sphingolipids are bioactive metabolites with well-established roles in human disease. Characterizing alterations in tissues with mass-spectrometry can reveal roles in disease etiology or identify therapeutic targets. However, the OCT-compound used for cryopreservation in biorepositories interferes with mass-spectrometry. We outline methods to analyze sphingolipids in human tissues embedded in OCT with LC-ESI-MS/MS.

Chemical Kinetics The reaction rate is the speed at which a chemical reaction occurs. The reaction rate is defined as the change in concentration of a component in the reaction with time. The speed of a reaction depends on several factors, including the concentration of reactants and the temperature at which the reaction is performed. Each reactant contributes to the speed of the reaction by a specific factor. This relationship is defined by the reaction rate law. Rate Law The rate law is an...

Source: Smaa Koraym at Johns Hopkins University, MD, USA Temperature Dependent Decomposition of Hydrogen PeroxideExpand We suggest that students work in pairs for this experiment. Equipment controls may vary. In this lab, you'll perform a decomposition reaction where a single compound breaks down into two or more simpler products. You will observe the decomposition of hydrogen peroxide into water and oxygen. This decomposition happens very slowly, so you will use iron (III) nitrate as a...
