All-trans-retinal

All-trans-retinal is a vitamin A-derived aldehyde and light-generated chromophore that plays a central role in vertebrate vision. When light strikes rhodopsin or a cone opsin, its bound 11-cis-retinal changes shape to all-trans-retinal, activating the photoreceptor and initiating visual signal transduction. Enzymes then reduce all-trans-retinal to all-trans-retinol, allowing transport through the visual cycle for regeneration of 11-cis-retinal in the retinal pigment epithelium. Studying this molecule helps explain photoreceptor function, vitamin A metabolism, and retinal disorders associated with impaired clearance or abnormal accumulation of retinaldehydes.

Subretinal injection has been widely applied in preclinical studies of stem cell replacement therapy for age-related macular degeneration. In this visualized article, we describe a less risky, reproducible and precisely modified subretinal injection technique via the trans-scleral approach to deliver cells into rat eyes.
