Antarctic Lake Sampling

Antarctic lake sampling is the systematic collection of water, sediment, and biological material from lakes in Antarctica to study life and environmental conditions in extreme, isolated ecosystems. Researchers access these environments through ice using carefully controlled water-collection or sediment-coring methods, while sterile equipment and sample preservation help maintain the original chemical and biological signals. The resulting samples support analyses of microbial communities, nutrient cycling, geochemistry, and ecological adaptation to cold, dark, or saline conditions. Antarctic lake sampling also provides evidence for past environmental change and helps assess how life might persist in other extreme environments, including extraterrestrial habitats.

Microbial eukaryotes are both a source of photosynthetically-derived carbon and top predatory species in permanently ice-covered Antarctic lakes. This report describes an enrichment culture approach to isolate metabolically versatile microbial eukaryotes from the Antarctic lake, Lake Bonney, and assesses inorganic carbon fixation potential using a radioisotope assay for Ribulose-1,5-bisphophate carboxylase oxygenase (RubisCO) activity.

A technique for laboratory estimation of net trophic transfer efficiency of polychlorinated biphenyl (PCB) congeners to piscivorous fish from their prey is presented. To maximize applicability of the laboratory results to the field, the piscivorous fish should be fed prey fish that are typically eaten in the field.

Laboratory determination of phosphorus release rates from sediment cores is a valuable approach for determining the role of internal phosphorus loading and guiding management decisions.

A novel sample holder for macromolecular X-ray crystallography along with a suitable handling protocol is presented. The system allows crystal growth, crystal soaking and in situ diffraction data collection at both, ambient and cryogenic temperature without the need of any crystal manipulation or mounting.
