Transient Transfection

Transient transfection is a laboratory method for temporarily introducing foreign nucleic acids into eukaryotic cells to produce a short-lived change in gene expression. DNA or RNA enters cells through approaches such as lipid-mediated delivery or electroporation, allowing the introduced material to direct protein production without becoming a stable part of the cell’s genome; expression typically decreases as the nucleic acid is degraded or diluted during cell division. In biology, transient transfection supports gene function studies, reporter assays, protein production, and evaluation of cellular responses to engineered genetic material. Its rapid, flexible design makes it useful for testing constructs before pursuing stable cell-line development.

Laboratory-scale production of eukaryotic proteins with appropriate post-translational modification represents a significant barrier. Here is a robust protocol with rapid establishment and turnaround for protein expression using a mammalian expression system. This system supports selective amino acid, selective labeling of proteins and small molecule modulators of glycan composition.

An Introduction to Transfection

2023

Transfection is the process of inserting genetic material, such as DNA and double stranded RNA, into mammalian cells. The insertion of DNA into a cell enables the expression, or production, of proteins using the cells own machinery, whereas insertion of RNA into a cell is used to down-regulate the production of a specific protein by stopping translation. While the site of action for transfected RNA is the cytoplasm, DNA must be transported to the nucleus for effective transfection. There, the...

Neuronal Transfection Methods

Transfection - the process of transferring genetic material into cells - is a powerful tool for the rapid and efficient manipulation of gene expression in cells. Because this method can be used to silence the expression of specific proteins or to drive the expression of foreign or modified proteins, transfection is an extremely useful tool in the study of the cellular and molecular processes that govern neuron function. However, mature neurons have a number of properties that make them...
