Basal Bodies

Basal bodies are microtubule-organizing structures that anchor cilia and flagella at the surface of many eukaryotic cells, helping establish cell polarity and movement. Derived from centrioles, each basal body contains a stable arrangement of microtubule triplets that nucleates the axonemal microtubules extending into a cilium or flagellum; associated proteins regulate its position, maturation, and attachment to the cell membrane. Basal bodies are essential for motile cilia, sensory cilia, and flagellar function in organisms ranging from protists to humans. Studying their assembly and defects helps explain developmental disorders, impaired mucociliary clearance, infertility, and other conditions linked to abnormal cilia.

This protocol describes the method of determining basal and forskolin-stimulated lipolysis in inguinal fat pads obtained from normal chow diet (NCD) or high fat diet (HFD) ± capsaicin fed wild type mice. As an index for lipolysis, glycerol release was measured from inguinal adipose fat pads.

Here we demonstrate our protocol for isolation of basal and submucosal gland duct cells from mouse tracheas. We also demonstrate the method of injecting stem cells into the dorsal mouse fat pad to create an in vivo model of submucosal gland regeneration.
