Cavefish Surface Fish

Cavefish and surface fish are contrasting forms of the Mexican tetra, Astyanax mexicanus, that provide a natural system for studying how environments shape animal development and evolution. In cave populations, persistent darkness and limited food have favored changes in embryonic eye development, pigmentation, metabolism, sensory systems, and behavior, whereas surface fish retain traits suited to sunlit habitats; these differences arise through heritable genetic variation and developmental regulation. Comparing the forms helps biologists link genes to traits, investigate adaptation and convergent evolution, and examine processes such as sleep, feeding, and tissue repair. This model supports broader research on evolution and human developmental disorders.

A series of basic methods to enable the study of the reproductive ecology of fish kept in aquaria are described. These are useful protocols for collecting fish using SCUBA, transporting live fish, and observing the reproductive behavior of wild-caught fish kept in aquaria.

When cells are placed in a hypotonic (low-salt) fluid, they can swell and burst. Meanwhile, cells in a hypertonic solution—with a higher salt concentration—can shrivel and die. How do fish cells avoid these gruesome fates in hypotonic freshwater or hypertonic seawater environments? Fish employ osmoregulatory strategies to balance bodily levels of water and dissolved ions (i.e., solutes), such as sodium and chloride. Imagine two solutions separated by a membrane that is permeable to water.

Morpholino Mediated Gene Editing: A Gene Knockdown Technique Using Translational Blocking Morpholino Oligonucleotides in Single-Celled Cavefish Eggs

This video describes a gene knockdown technique using translational blocking morpholinos microinjected in the one-cell stage of the cavefish. Morpholinos are DNA oligonucleotides that bind to the complementary sequence of target mRNA and block its translation to a protein. Consequently, morpholinos are used as molecular tools to stop or reduce a gene’s expression transiently.

Manipulation of Gene Function in Mexican Cavefish

We describe approaches for the manipulation of genes in the evolutionary model system Astyanax mexicanus. Three different techniques are described: Tol2-mediated transgenesis, targeted manipulation of the genome using CRISPR/Cas9, and knockdown of expression using morpholinos. These tools should facilitate the direct investigation of genes underlying the variation between surface- and cave-dwelling forms.
