Cell Holograms

Cell holograms are three-dimensional representations of cells created by recording and reconstructing the light waves that pass through or scatter from cellular structures. In digital holographic microscopy, a laser or other coherent light source produces an interference pattern between an object beam and a reference beam; computational reconstruction then converts this pattern into quantitative maps of cell shape, thickness, and optical phase without requiring fluorescent labels. In biology, cell holograms support noninvasive analysis of morphology, growth, movement, and viability over time. They can help researchers monitor living cells, assess responses to treatments, and study dynamic cellular processes while minimizing phototoxicity and preparation artifacts.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.
