Clonal Proliferative Potential

Clonal proliferative potential is the capacity of an individual cell to undergo repeated divisions and generate a population of genetically related descendants, making it a key property in biology. It depends on the cell’s ability to maintain self-renewal while preserving viability and, in some contexts, producing differentiated progeny; clonogenic assays and single-cell lineage tracing can measure this capacity. Assessing clonal proliferative potential helps characterize stem and progenitor cells, compare tumor cell behavior, and evaluate tissue regeneration or drug responses. These analyses connect cell-level division patterns with lineage commitment, heterogeneity, and long-term tissue maintenance.

Although proliferative polyploid cells are necessary to analyze chromosomal instability of polyploid cells, creating such cells from nontransformed human cells is not easy. The present report describes relatively simple procedures to establish proliferative tetraploid cells free of a diploid population from normal human fibroblasts.
