Cytoplasmic Mrna Removal

Cytoplasmic mRNA removal is the regulated degradation of messenger RNA in the cell cytoplasm, controlling how long transcripts remain available for protein synthesis. It commonly begins with deadenylation, which shortens the poly(A) tail, followed by decapping and exonucleolytic digestion from either the 5′ or 3′ end; endonucleolytic cleavage and RNA surveillance pathways can also accelerate clearance of faulty or unnecessary transcripts. By adjusting mRNA stability, cells tune gene expression, eliminate defective messages, and respond to developmental or environmental signals. Studying this process clarifies post-transcriptional regulation and how its disruption can contribute to disease.

Regulated mRNA Transport

2020

In eukaryotes, transcription and translation are compartmentalized; an mRNA is first synthesized in the nucleus and then selectively transported to the cytoplasm for protein synthesis. Before transport, a pre-mRNA undergoes several steps of post-transcriptional modifications including splicing, 5' capping, and the addition of a poly-adenine tail. Various proteins bind to the pre-mRNA during these modifications. The mRNA transport takes place with the help of multiple proteins playing specific...

Nuclear Export of mRNA

Before mRNAs are exported to the cytoplasm, it is crucial to check each mRNA for structural and functional integrity. Eukaryotic cells use several different mechanisms, collectively known as mRNA surveillance, to look for irregularities in mRNAs. Irregular or aberrant mRNA are rapidly degraded by various enzymes. If a defective mRNA escapes the surveillance, it would be translated into a protein which would either be non-functional or not function properly. One of the primary irregularities in...

The cytoplasm consists of organelles and a framework of protein scaffolds called the cytoskeleton suspended in an aqueous solution, the cytosol. The cytosol is a rich broth of water, ions, salts, and various organic molecules.Protein Folding and MisfoldingThe cytoplasm is the location for several cellular processes, including protein synthesis and folding. The aqueous nature of the cytosol promotes protein folding such that the hydrophobic amino acid side chains are buried in the protein core...

mRNA Stability and Gene Expression

2020

The structure and stability of mRNA molecules regulates gene expression, as mRNAs are a key step in the pathway from gene to protein. In eukaryotes, the half-life of mRNA varies from a few minutes up to several days. mRNA stability is essential in growth and development. The absence of the proteins regulating its stability, such as tristetraprolin in mice, can cause systemic issues, including bone marrow overgrowth, inflammation, and autoimmunity.
