Ductal Obstruction

Ductal obstruction is the partial or complete blockage of a biological duct, preventing the normal passage of fluids, secretions, or cells through a tubular pathway. In exocrine systems, obstruction can result from accumulated material, swelling, scarring, or a mass that narrows the duct and causes fluid to build up upstream, disrupting tissue function. Studying ductal obstruction helps explain disorders of the bile ducts, pancreas, salivary glands, kidneys, and reproductive tract, while supporting the interpretation of symptoms, imaging findings, and laboratory results. Understanding its mechanisms also informs research into inflammation, tissue injury, and treatments that restore drainage or relieve pressure.

2023

This video describes the technique of differentiating pancreatic acinar cells to ductal cells in ex vivo 3D culture. This helps to understand the mechanisms regulating the acinar-to-ductal metaplasia (ADM), a key process in the early development of pancreatic cancer.

Unilateral Ureteral Obstruction Murine Model: A Surgical Procedure to Obstruct the Urinary Flow from a Kidney in Mouse Models

This video demonstrates the procedure to generate a unilateral ureteral obstruction mouse model. The procedure involves cutting one of the ureters of the mouse to obstruct urine flow.

Reprogramming Pancreatic Ductal Adenocarcinoma to Pluripotency

2024

The present protocol describes the reprogramming of Pancreatic Ductal Adenocarcinoma (PDAC) and normal pancreatic ductal epithelial cells into induced pluripotent stem cells (iPSCs). We provide an optimized and detailed, step-by-step procedure, from preparing lentivirus to establishing stable iPSC lines.

The murine model of irreversible unilateral ureteric obstruction (UUO) is presented together with the model of reversible UUO in which the ureteric obstruction is relieved by anastomosis of the severed ureter into the bladder. These models enable the study of renal inflammation and scarring as well as tissue remodeling.

Ultrasound-Guided Implantation: A Technique to Implant Pancreatic Ductal Adenocarcinoma Cells into an Adult Mouse
