Epithelial Barrier

An epithelial barrier is a specialized layer of epithelial cells that separates tissue compartments, controls exchange with the environment, and protects underlying tissues from physical, chemical, and microbial threats. Its function depends on coordinated cell polarity and junctional structures, including tight junctions that regulate paracellular passage and adherens junctions and desmosomes that maintain cohesion under mechanical stress. In biology, epithelial barriers are studied in the intestine, skin, lungs, and kidney to understand selective permeability, transport, inflammation, and tissue repair. Barrier models and permeability assays also support research into disease mechanisms, drug delivery, toxicology, and therapeutic strategies that restore barrier integrity.

Current treatments of inflammatory bowel diseases (IBD) aim at alleviating disease symptoms but may cause severe side effects. Thus, alternative strategies are being investigated in animal colitis models. Here, we explain how the beneficial effects of diet supplements on clinical IBD signs are analyzed in such a colitis model.

The epithelial cells of the choroid plexus (CP) form the blood-cerebrospinal fluid barrier (BCSFB). An in vitro model of the BCSFB employs human choroid plexus papilloma (HIBCPP) cells. This article describes culturing and basolateral infection of HIBCPP cells using a cell culture filter insert system.
