Fixation Permeabilization

Fixation permeabilization is a sample-preparation method in biology that preserves cellular structures while allowing probes and antibodies to enter cells. Fixation stabilizes proteins and membranes, often through chemical cross-linking, whereas permeabilization creates pores by partially disrupting lipid membranes with detergents or organic solvents. Together, these steps enable immunofluorescence staining, intracellular antigen detection, and microscopic analysis of cell organization. Their conditions must be carefully controlled because excessive fixation can mask epitopes, while harsh permeabilization can damage cellular morphology or remove soluble components. Optimized treatment supports reliable imaging, localization studies, and quantitative analysis in cell biology and biomedical research.

Two basic types of preparation are used to visualize specimens with a light microscope: wet mounts and fixed specimens. The simplest type of preparation is the wet mount, in which the specimen is placed in a drop of liquid on the slide. A liquid specimen can be directly deposited on the slide using a dropper. Fluorescence in-situ hybridization (FISH) is often required in combination with histopathology and molecular diagnostics for selection of therapy in personalized medicine. A novel non-cross-linking, formalin-free tissue fixative that allows high quality morphologic, molecular and FISH analyses from the same specimen by addition of a post-fixation step before FISH is presented.
