Fluorescent Labeling

Fluorescent labeling is a method for attaching fluorescent molecules to biological targets so they can be detected and located through emitted light. When illuminated at a specific excitation wavelength, a fluorophore absorbs energy and releases it at a longer emission wavelength, allowing labeled cells, proteins, nucleic acids, or other structures to be distinguished from their surroundings. In biology, researchers use fluorescent dyes, proteins, and antibody conjugates for microscopy, cell tracking, gene expression analysis, and protein localization. Careful selection of labels, fixation or live-cell conditions, and imaging settings helps improve signal specificity while limiting photobleaching and background fluorescence.

Here we describe a basic protocol for fluorescent labeling of different elements of heart tubes from larva and adult Drosophila melanogaster. These specimens are well-suited for imaging via fluorescent or confocal microscopy. This technique permits detailed structural analysis of the features of the hearts from a powerful model organism.
