Fluorescent Marker Expression

Fluorescent marker expression is the production of fluorescent proteins or other fluorescent labels in cells, enabling researchers to visualize specific structures, molecules, or biological activities. In genetic systems, a DNA sequence encoding a fluorescent protein is introduced or linked to a regulatory element, causing the marker to be produced in selected cells or under particular conditions; excitation light then induces the protein to emit detectable fluorescence. This approach supports microscopy-based analysis of gene expression, cell identity, protein localization, and dynamic cellular processes. In biology, fluorescent markers help track development, investigate disease mechanisms, and evaluate experimental changes over time.

Here we present a Fluorescence Activated Cell Sorting (FACS) protocol to study molecular alterations in Fos-expressing neuronal ensembles from both fresh and frozen brain tissue. The use of frozen tissue allows FACS isolation of many brain areas over multiple sessions to maximize the use of valuable animal subjects.
