Gel-assisted Swelling

Gel-assisted swelling is a biological imaging technique that enlarges embedded specimens to reveal molecular and cellular structures with greater spatial detail. In this approach, biomolecules are anchored within a polymer hydrogel, the surrounding tissue is homogenized or softened, and the gel absorbs water, expanding the specimen while preserving its relative organization. This physical magnification can improve the study of protein localization, cellular architecture, and tissue morphology, supporting research in developmental biology, neuroscience, and disease biology. By making nanoscale relationships more accessible with conventional microscopy, gel-assisted swelling complements advanced imaging methods and helps connect molecular organization to biological function.

Forming Giant-sized Polymersomes Using Gel-assisted Rehydration

We present a protocol to rapidly form giant polymer vesicles (pGVs). Briefly, polymer solutions are dehydrated on dried agarose films adhered to coverslips. Rehydration of the polymer films results in rapid formation of pGVs. This method greatly advances the preparation of synthetic giant vesicles for direct applications in biomimetic studies.
