Total Protein Stain

Total protein stain is a laboratory technique that visualizes the overall protein content in a biological sample, providing a rapid way to assess protein loading and separation. After electrophoresis or transfer to a membrane, dyes such as Coomassie Brilliant Blue, Ponceau S, or fluorescent stains bind protein residues and produce visible bands whose intensity reflects relative protein abundance. In biology, total protein staining helps evaluate sample quality, confirm efficient transfer, identify uneven loading, and normalize Western blot signals without relying exclusively on a single housekeeping protein. It supports more reliable comparisons in studies of cell structure, expression, and biochemical responses.

The stain-free technology allows researchers to visualize protein separation quality, to verify the transfer efficiency, and most importantly, to validate the change in the protein of interest using total protein quantification as a reliable loading control.

Members of the Burkholderia genus are pathogens of clinical importance. We describe a method for total bacterial protein extraction, using mechanical disruption, and 2-D gel electrophoresis for subsequent proteomic analysis.
