Head Kidney Exposure

Head kidney exposure refers to the contact of the head kidney, a major hematopoietic and immune organ in fish, with environmental chemicals, pathogens, or other experimental agents. During exposure, substances absorbed from water or food can reach head-kidney tissue and alter resident immune cells, blood-cell production, gene expression, or inflammatory signaling. Researchers use this model to evaluate immunotoxicity, environmental stress, disease responses, and the effects of pollutants on fish health. Measuring cellular, molecular, and physiological changes in the head kidney can reveal early biological impacts and help connect contaminant exposure with impaired immunity, altered hematopoiesis, or reduced organismal fitness.

In this video, we demonstrate the depigmentation of adult zebrafish kidney and its subsequent preparation for staining procedures. Post-staining, the adult zebrafish kidney can be used for studying kidney regeneration, phenotypic characterization of renal disease models, and nephron functionality.

Dissection of the Adult Zebrafish Kidney

The zebrafish kidney is home to both renal and hematopoietic adult stem/progenitor cells, and represents an outstanding opportunity to study these cell types and their progeny in a vertebrate model organism. Here, we demonstrate a detailed dissection procedure that enables the researcher to identify and surgically remove the adult zebrafish kidney, which can be used for applications such as cell isolation, transplantation, and expression studies of kidney and/or blood cell populations.

The severe organ shortage has resulted in increased use of marginal kidney grafts for transplantation. This has triggered interest in alternative storage methods, since marginal grafts especially tolerate cold storage poorly. The technique of normothermic ex vivo kidney perfusion (NEVKP) represents a novel preservation method for kidney grafts prior to transplantation.
