Heterozygous

Heterozygous describes a genetic state in which an individual carries two different alleles of a gene at the same locus, one inherited from each parent. In diploid organisms, these alleles occupy corresponding positions on homologous chromosomes and may interact through patterns such as dominance, incomplete dominance, or codominance, influencing which traits are expressed. Heterozygous genotypes are central to understanding inheritance, genetic variation, and population diversity. They also help explain carrier status for recessive conditions, predict offspring outcomes using genetic crosses, and interpret how specific variants contribute to biological traits or disease risk.

2023

Novel strategies to faithfully model somatic mutations in hematopoietic stem and progenitor cells (HSPCs) are necessary to better study hematopoietic stem cell biology and hematological malignancies. Here, a protocol to model heterozygous gain-of-function mutations in HSPCs by combining the use of CRISPR/Cas9 and dual rAAV donor transduction is described.

Probability Laws

Overview The probability of inheriting a trait can be calculated using the sum and product rules. The sum rule is used to calculate the probability of mutually exclusive events. The product rule predicts the probability of multiple independent events. These probability rules determine theoretical probability—the likelihood of events occurring before they happen. Empirical probability, by contrast, is calculated based on events that have already occurred. The Sum and Product Rules Are...

An Allele-specific Gene Expression Assay to Test the Functional Basis of Genetic Associations

2010

Genetic associations often remain unexplained at a functional level. This method aims to assess the effect of phenotype-associated genetic markers on gene expression by analyzing cells heterozygous for transcribed SNPs. The technology allows accurate measurement by MALDI-TOF mass spectrometry to quantify allele-specific primer extension products.

Assessing Social Dominance in Mouse Models Using the Tube Test

This protocol describes a behavioral assay to evaluate social dominance in rodents using the tube test. Social dominance remains stable over time, and several models of developmental and neurological disorders exhibit robust social dominance abnormalities. Therefore, the tube test serves as a convenient outcome measure for mechanistic studies or preclinical therapeutic screening.

2026

To model how inflammatory signals influence neurodevelopment in genetically susceptible organisms, we present a protocol for microinjecting IL-6 into the duct of Cuvier (common cardinal vein) of zebrafish larvae, enabling controlled induction of inflammation and assessment of its developmental impact in the context of tuberous sclerosis complex (TSC).
