Alzheimer’s disease (AD) is a progressive and multifactorial neurodegenerative disorder with a substantial and growing global health burden. Advances in disease biology, biomarkers, and therapeutic development have accelerated the emergence of disease-modifying approaches, including amyloid-targeting therapies, while expanding research toward tau, neuroinflammation, synaptic dysfunction, metabolic disturbances, vascular mechanisms, and other pathogenic processes.
Despite these advances, major challenges remain, including the complex and heterogeneous pathophysiology of AD, incomplete understanding of disease mechanisms, limited translation from preclinical models to patients, blood–brain barrier constraints, treatment-related adverse effects, difficulties in early diagnosis and patient selection, biomarker validation, therapeutic resistance, and the need for effective interventions across different stages of disease.
This Collection aims to provide a comprehensive platform for recent advances in therapeutic approaches or drug development for AD, encompassing the discovery and validation of therapeutic targets, development and optimization of candidate interventions, evaluation of therapeutic efficacy and safety, and translation of promising strategies from preclinical research into clinical applications. Contributions exploring emerging therapeutic approaches, drug-development strategies, various intervention modalities, mechanisms of therapeutic action, drug delivery and pharmacological evaluation, biomarker-guided treatment, and clinical development are encouraged.
Methods articles, original research articles, review articles, and case reports are welcome to capture diverse advances across the preclinical and clinical spectrum of AD therapeutic research. By bringing together methodological innovations, experimental findings, comprehensive evidence, and clinically relevant developments, this Collection will facilitate knowledge exchange across disciplines, help researchers identify emerging opportunities and unresolved challenges, and support the translation of promising therapeutic discoveries into clinically meaningful interventions.
