In Vitro Cellular Mri

In vitro cellular MRI is an imaging approach that uses magnetic resonance signals to visualize and characterize cells maintained under laboratory conditions, providing a non-destructive way to study cellular behavior and labeling. Cells may be incubated with MRI contrast agents, such as iron oxide or gadolinium-based probes, which alter local relaxation times, especially T1 or T2, and create measurable signal differences. Researchers use the technique to track labeled cell populations, evaluate probe uptake and retention, and examine treatment effects in controlled culture systems. These measurements support cell biology, regenerative medicine, drug development, and refinement of cellular imaging methods before animal or clinical studies.

Early detection of apoptosis may identify at-risk cell populations in a variety of diseases. Here we demonstrate a method to link an early apoptosis-detection protein (Annexin V) to a MRI-detectable iron oxide nanoparticle (SPIO). This method may be extended to other proteins of interest to generate MRI-detectable molecular imaging probes.

An In Vitro Assay to Measure Antibody-Dependent Cellular Phagocytosis by Phagocytes

This video demonstrates an assay to measure the antibody-dependent cellular phagocytosis (ADCP) by breast milk phagocytes. Fluorescently labeled microspheres conjugated to an antigen-antibody complex, are combined with a cell suspension isolated from breast milk to induce antibody-dependent cellular phagocytosis by the phagocytic leukocytes. Upon labeling the cells with antibodies against common leukocyte antigen (CD45), flow cytometry is used to measure the fluorescence intensity of...

The protocol presents two methods to determine the kinetics of the fluorogenic RNA aptamers Spinach2 and Broccoli. The first method describes how to measure fluorogenic aptamer kinetics in vitro with a plate reader, while the second method details the measurement of fluorogenic aptamer kinetics in cells by flow cytometry.
