Lineage-specific Markers

Lineage-specific markers are measurable molecules or traits that identify cells belonging to a particular developmental, tissue, or cellular lineage, making them essential for distinguishing cell identity and fate. They typically consist of characteristic proteins, transcripts, or surface antigens whose expression is regulated as progenitor cells differentiate and maintain specialized functions. In biology, researchers use marker panels with methods such as immunostaining, flow cytometry, and gene-expression analysis to track differentiation, isolate cell populations, and assess developmental relationships. These markers support studies of embryogenesis, stem cell biology, tissue regeneration, disease progression, and cancer, while combinations of markers can improve identification when no single feature uniquely defines a lineage.

Visualization and Quantification of Mesenchymal Cell Adipogenic Differentiation Potential with a Lineage Specific Marker

Traditional methods of assessing adipogenic differentiation are cheap and easy to use, but are not specific to changes in gene expression. We have developed an assay to quantify mesenchymal cell differentiation into mature adipocytes using a lineage specific marker. This assay has diverse applications across basic research and clinical medicine.

Co-localization of Cell Lineage Markers and the Tomato Signal

We developed two sets of tracing combinations in Rosa26tdtomato (ubiquitously expressed in all cells)/Cre (specifically expressed in chondrocytes) mice: one with 2.3Col1a1-GFP (specific to osteoblasts) and one with immunofluorescence (specific to bone cells). The data demonstrate the direct transformation of chondrocytes into bone cells.

Genome editing tools such as the CRISPR (Clustered Regularly Interspaced Short Palindromic Repeats)/Cas (CRISPR-associated) system have greatly improved gene targeting efficiency in human induced pluripotent stem cells (hiPSCs). This manuscript describes a protocol for generating lineage specific hiPSC reporter using CRISPR/Cas system assisted homologous recombination.

We describe use of ImageStream technology (www.amnis.com), which combines quantitative flow cytometry with simultaneous high-resolution digital imaging, to quantify cellular mechanisms of primary immune cells from well-defined patient cohorts. Our studies provide a blueprint for translational investigations to quantify lineage specific cellular responses in small samples from subject cohorts.

Extraction, Labeling, and Purification of Lineage-Specific Cells from Human Antral Follicles

Here, we present protocols for the identification and purification of ovarian cells from antral follicles. We elaborate on methods for processing whole ovaries for the cryopreservation of cortical strips while also harvesting intact antral follicles that are treated enzymatically to liberate multiple follicle resident cell types, including granulosa, theca, endothelial, hematopoietic, and stromal cells.
