Meiotic Anther Fixation

Meiotic anther fixation is a tissue-preservation method used to maintain the cellular and chromosomal features of developing pollen during meiosis, enabling accurate microscopic analysis. Chemical fixatives rapidly stop cellular activity and stabilize tissue components, preserving chromosome structure and the spatial organization of meiotic cells as anthers undergo processing, staining, and sectioning or squashing. In plant biology, this technique supports identification of meiotic stages, analysis of chromosome pairing and segregation, and evaluation of pollen development and fertility. Reliable fixation provides essential material for plant cytogenetics, reproductive research, breeding programs, and investigations of abnormalities that can affect seed production.

Mammalian meiosis can be examined by utilizing a technique to prepare meiotic chromosome spreads. Here, we demonstrate a method of preparing surface-spread nuclei from mouse spermatocytes.

Two basic types of preparation are used to visualize specimens with a light microscope: wet mounts and fixed specimens.
