MOV10 and MOV10L1 are RNA helicase proteins that regulate RNA metabolism, making them important for post-transcriptional gene control and germ-cell development. Using ATP-dependent helicase activity and RNA binding, MOV10 participates in RNA remodeling and regulation, whereas MOV10L1 acts in germ cells to bind piRNA precursor transcripts and support their processing into PIWI-associated small RNAs. These proteins help control gene expression, transposon silencing, and genome stability, with MOV10L1 being especially important for meiosis and male fertility. Studying their molecular functions clarifies how RNA-based pathways maintain cellular identity and protect reproductive genomes.

In this video, we demonstrate the procedure to determine the helicase activity of a target protein to unwind the biotin-labeled dsRNA substrate. The activity of the enzyme was identified by analyzing the electrophoretic mobility shift, followed by a chemiluminescence assay using chemiluminescent enzyme-conjugated streptavidin.
