Intracellular Sodium Measurement

Intracellular sodium measurement quantifies the concentration or dynamics of Na⁺ within cells, providing insight into ion homeostasis, membrane transport, and cellular signaling. In many biological studies, sodium-sensitive fluorescent indicators enter cells, bind Na⁺, and change their fluorescence intensity or spectral properties; microscopy and calibration then convert these signals into intracellular sodium estimates. This approach helps assess the activity of ion channels, pumps, and transporters, as well as changes caused by osmotic stress, electrical stimulation, or disease-related dysfunction. Measuring sodium at the cellular level supports research in physiology, neuroscience, pharmacology, and cell biology.

Single Cell Measurements of Vacuolar Rupture Caused by Intracellular Pathogens

We describe a method for tracking the endomembrane rupture elicited by the intracellular bacteria Shigella flexneri and Mycobacterium tuberculosis upon host cell invasion. Our assay makes use of CCF4, a host cytoplasmic FRET probe in live or fixed cells. This reporter is degraded by an enzyme activity present on the bacterial surface.

In this protocol a method to measure intracellular protein refolding after heat shock is described. This method can be used to study foldases like molecular chaperones and their co-factors or compounds able to influence their activity. Firefly luciferase activity is used as reporter to measure chaperone refolding activity.
