Neutrophil Activation

Neutrophil activation is the process by which these innate immune cells detect threats and rapidly respond to infection or tissue injury, making it a central component of inflammation. Recognition of pathogen- or damage-associated molecular patterns by cell-surface and intracellular receptors triggers signaling pathways that promote chemotaxis, adhesion, phagocytosis, degranulation, reactive oxygen species production, and, in some conditions, formation of neutrophil extracellular traps. These coordinated responses help eliminate microbes and shape the surrounding immune reaction. Studying neutrophil activation supports research into host defense, sepsis, chronic inflammatory diseases, autoimmune disorders, and therapies designed to strengthen antimicrobial activity while limiting tissue damage.

Peritoneal Low-density Neutrophil Isolation: A Technique to Obtain Low-density Neutrophils from Peritoneal Lavage Fluid Using Magnetic Activated Cell Sorting

2023

In this video, we describe the procedure for isolating low-density neutrophils, or LDNs, from the peritoneal lavage fluid of a postoperative human patient. The LDNs thus obtained can be used in further downstream experiments to understand their role in tumor growth and progression.

An In Vitro Assay for Measuring Neutrophil Serine Protease Activity Using a Fluorescent Reporter

2025

This video demonstrates an in vitro assay to quantify neutrophil serine protease activity in sputum samples. The sample containing secreted protease is incubated with a fluorescent reporter bearing a recognition motif. Cleavage of the motif by the protease enables individual fluorescence emission by the donor and acceptor fluorophore of the reporter, indicating protease activity in the sample.

Induction and Visualization of Neutrophil Extracellular Traps in Pre-Labeled Neutrophils

2025

This video demonstrates the induction and visualization of neutrophil extracellular traps or NETs in pre-labeled neutrophils using patient serum containing anti-neutrophil cytoplasmic antibodies. Furthermore, using an impermeable DNA dye and fluorescence microscopy enables the visualization of NETs.
