Human Oral Epithelium

Human oral epithelium is the stratified squamous tissue that lines the mouth, where it forms a protective interface between underlying tissues and the external environment. It withstands mechanical stress, limits water loss, and helps block microbes while supporting sensation and local immune defense. In its basal layer, cells proliferate and migrate toward the surface, progressively differentiating into keratinized or nonkeratinized layers; cell junctions and surface lubrication maintain barrier integrity. Studying human oral epithelium supports research on mucosal wound healing, oral infections, cancer, and interactions with dental materials and therapeutic formulations. Its organization and regenerative behavior also provide models for epithelial biology and tissue engineering.

The following protocol describes a novel method for chronic oral drug administration using an orally dissolving strip (ODS) in lieu of the more commonly used oral gavage method. We demonstrate that preclinical, oral drug delivery using the ODS method represents a safe, convenient, and humane alternative to oral gavage.
