Onion Cell Preparation

Onion cell preparation is a basic microscopy technique that isolates and mounts onion epidermal cells so their structures can be observed, making it a useful introduction to plant cell biology. A thin inner epidermal layer is peeled from the onion, placed on a microscope slide in a liquid mount, and often stained to increase contrast; the cells’ large vacuoles, cell walls, and nuclei can then become distinguishable under magnification. This preparation helps students connect cell theory with direct observation while providing a simple model for studying plant-cell organization, microscopy, staining, and the effects of mounting conditions.
