Proximal Colon Microbiota

Proximal colon microbiota refers to the community of microorganisms inhabiting the first section of the large intestine, where microbial activity strongly influences intestinal biology. In this oxygen-limited environment, bacteria metabolize undigested carbohydrates and other intestinal contents through fermentation, producing short-chain fatty acids and interacting with the mucosal surface and host immune system. Studying these microbial communities helps researchers understand nutrient processing, epithelial function, and host-microbe interactions in biology. Characterizing their composition and metabolic outputs also supports investigation of how changes in the proximal colon may relate to gastrointestinal health, disease mechanisms, and responses to diet or other interventions.

In this video, we describe a surgical procedure to generate mouse proximal colon loop or pcLoop segment followed by intraluminal injection of fluorescent markers into the segment to study the intestinal epithelial permeability.

An Ex Vivo Gut Organ Culture System to Study Host-Microbiota Interactions

In this video, we describe the methodology for setting up an ex vivo gut organ culture system to study the interactions between mouse intestinal tissue fragments and specific gut bacteria. The device consists of two pairs of input-output ports, the first connecting the intestinal tissue lumen and supplying the specific bacterial suspension into it, and the second continuously supplying media to the device's wells to maintain ex vivo tissue viability.

Begin with a rat model with sutures tied around its upper molar teeth to support bacterial colonization.Repeatedly smear the sutures with a gum disease-causing oral pathogenic bacterium using a gel-like solution. The bacteria colonize the gum around the sutures.Colonization triggers inflammation and damages the tissues. The bacteria release toxins that diffuse through damaged tissues and enter the bloodstream. These circulating toxins reach the gut, cause inflammation, disrupt the epithelial...

Evaluating the Effects of Fecal Microbiota Transplantation in an Interleukin-10 Deficient Mouse
