Thallium Flux Assay

The thallium flux assay is a cell-based method for measuring potassium channel activity by tracking the movement of thallium ions across cell membranes. Because thallium ions can pass through many potassium-selective pathways, cells are loaded with a thallium-sensitive fluorescent indicator, and changes in fluorescence reveal ion influx after channel activation or membrane stimulation. This assay provides a rapid, quantitative readout of channel function and inhibition in living cells. It is widely used in biology and pharmacology to characterize ion-channel mechanisms, compare compound potency, and screen potential therapeutics targeting potassium channels.

Extracellular Flux Assay: A Method for Measuring Metabolic Profile of Cells

2023

This protocol provides a biochemical approach for measuring extracellular acidification rate (ECAR) and oxygen consumption rate (OCR) of primary leukemic cells with extracellular flux assay. The assessment of the metabolic profile of these cells helps to characterize their demands, leading to personalized therapy.

Monitoring B Cell Activation with Antigenic Liposomes using a Calcium-Flux Assay

2025

This video showcases the assessment of B cell activation through the utilization of antigenic liposomes. B cells treated with calcium-sensitive Indo-1 dye are distinguished using flow cytometry, and their violet-to-blue fluorescence ratio is analyzed. As the cells encounter antigenic liposomes, it triggers B cell activation through calcium influx, progressively enhancing the violet-to-blue fluorescence signal ratio over time.

This protocol describes the use of the Ramsay assay to measure fluid secretion rates from isolated Malpighian (renal) tubules from Drosophila melanogaster. In addition, the use of ion-specific electrodes to measure sodium and potassium concentrations in the secreted fluid, allowing calculation of transepithelial ion flux, is described.

Electric Flux

The concept of flux describes how much of something goes through a given area. More formally, it is the dot product of a vector field within an area. For a better understanding, consider an open rectangular surface with a small area that is placed in a uniform electric field. The larger the area, the more field lines go through it and, hence, the greater the flux; similarly, the stronger the electric field (represented by a greater density of lines), the greater the flux. On the other hand, if...
