Tx-114 Surfactant

Tx-114 surfactant is a nonionic detergent used in biology to solubilize membrane-associated molecules and separate them from other cellular components. Its key property is temperature-dependent phase separation: above its cloud point, an aqueous solution divides into detergent-rich and detergent-poor phases, allowing hydrophobic proteins, lipids, or membrane fragments to partition selectively. Researchers use Tx-114 for membrane protein extraction, biochemical fractionation, and the removal of lipids or detergent-soluble contaminants from biological samples. Because phase separation can preserve protein complexes under relatively mild conditions, this surfactant supports studies of membrane composition, protein localization, and cellular biochemistry.

2016

Repeated pulmonary lavages in anesthetized pigs induce lung injury resembling major aspects of human acute respiratory distress syndrome (ARDS). For this purpose the lungs are repeatedly lavaged with 0.9% saline at 37 °C. The goal of the protocol is a reproducible mitigation of gas exchange and hemodynamics for research in ARDS.

This paper demonstrates the experimental procedure to measure terminal settling velocities of spherical particles in surfactant-based shear thinning viscoelastic fluids. Fluids over a wide range of rheological properties are prepared and settling velocities are measured for a range of particle sizes in unbounded fluids and fluids between parallel walls.

(2018)This video demonstrates the enrichment of bacterial lipoproteins using a non-ionic detergent that undergoes phase separation upon warming. Through sequential temperature shifts and centrifugation steps, lipoproteins are selectively partitioned into the detergent phase, while non-lipoprotein contaminants...
