Type Iv Secretion System

The Type IV secretion system is a multiprotein transport apparatus used by bacteria to move DNA, proteins, and other effector molecules into neighboring cells or host cells. Its membrane-spanning components assemble into a secretion channel and often a pilus, using energy from ATPases to drive substrate transfer across the bacterial envelope. In biology, Type IV secretion systems explain bacterial conjugation, horizontal gene transfer, and key mechanisms of host-cell manipulation during infection. Studying these systems supports research on antibiotic resistance, microbial evolution, pathogen virulence, and strategies for developing antimicrobial therapies or engineered delivery platforms.

Here we describe a method to visualize the oncogenic bacterial organelle known as the Cag Type IV Secretion System (Cag-T4SS). We find that the Cag-T4SS is differentially produced on the surface of H. pylori in response to varying conditions of iron availability.

Stent implants in stenosed arterial curvatures are prone to "Type IV" failures involving the complete transverse fracture of stents and linear displacement of the fractured parts. We present a protocol for detection of secondary flow (vortical) structures in a curved artery model, downstream of clinically relevant "Type IV" stent failures.
