Vacuum Sealing

Vacuum sealing is a packaging and preservation method that removes air from a container or flexible barrier before creating an airtight seal. The process lowers oxygen exposure and limits moisture exchange, which can slow oxidation, desiccation, and the growth of many aerobic microorganisms, although it does not sterilize biological materials. In biology, vacuum-sealed packaging can help protect specimens, reagents, culture components, foods used in laboratory studies, and other moisture-sensitive materials during storage or transport. Its effectiveness depends on material compatibility, seal integrity, temperature, and appropriate refrigerated or frozen conditions.

The present protocol describes the maceration and cleaning of cadaveric bone with a vacuum-sealed, hot water bath immersion technique. This is a low-cost, safe, and effective method to produce anatomical specimens for surgical planning and medical education as an alternative to three-dimensional (3D) printed models.

Key procedures to optimize the sealing process and achieve real-time monitoring of the metal-to-glass seal (MTGS) structure are described in detail. The embedded fiber Bragg grating (FBG) sensor is designed to achieve online monitoring of temperature and high-level residual stress in the MTGS with simultaneous environmental pressure monitoring.

A novel sealing protocol to produce water-tight silicone-based seals for membranes of Polydimethylsiloxane (PDMS) chip devices. Intended for labs setting-up chip models and small-scale chip platform development. We demonstrate the protocol using plastic and native tissue membranes. This procedure only requires PDMS, toluene, mold, membrane, and a vacuum chamber.
