Virus Titration Method

Virus titration methods are laboratory procedures used to measure the concentration of infectious virus in a sample, an essential step for comparing experiments and standardizing biological assays. Typically, a virus suspension is serially diluted and added to susceptible cells, where infection produces visible plaques, cytopathic effects, or another defined endpoint; the dilution yielding a countable response is used to calculate infectious titer. In biology research, virus titration supports vaccine and antiviral testing, viral growth studies, diagnostic assay development, and controlled infection experiments. Reliable titers improve reproducibility by enabling researchers to apply consistent infectious doses across samples and conditions.

Generation, Amplification, and Titration of Recombinant Respiratory Syncytial Viruses

We describe a method for generating and amplifying genetically modified respiratory syncytial viruses (RSVs) and an optimized plaque assay for RSVs. We illustrate this protocol by creating two recombinant viruses that respectively allow quantification of RSV replication and live analysis of RSV inclusion bodies and inclusion bodies-associated granules dynamics.
