4-hydroxy-tamoxifen

4-Hydroxy-tamoxifen (4-OHT) is a biologically active metabolite of tamoxifen that binds estrogen receptors and is widely used to study hormone-responsive cancer. Its structure enables high-affinity receptor binding, allowing it to block estrogen-dependent transcription in breast tissue while producing tissue-specific estrogen-like effects elsewhere. In cancer research, 4-OHT helps model estrogen receptor signaling and evaluate therapeutic responses in estrogen receptor-positive breast cancer cells and tumors. It is also used to activate tamoxifen-inducible CreERT2 recombinase, enabling controlled gene activation or deletion in experimental cancer models. These applications connect drug action with mechanisms of tumor growth and treatment resistance.

We have synthesized a novel analogue of pancratistatin with comparable anti-cancer activity as native pancratistatin; interestingly, combinatory treatment with tamoxifen yielded a drastic enhancement in apoptotic and autophagic induction by mitochondrial targeting with minimal effect on noncancerous fibroblasts. Thus, JCTH-4 in combination with tamoxifen could provide a safe anti-cancer therapy.

Dipeptidyl Peptidase 4 Inhibitors

2024

Dipeptidyl peptidase 4 (DPP-4) is a serine protease widely distributed in the body. It's involved in the inactivation of GLP-1 and GIP hormones, which are crucial for insulin regulation. DPP-4 inhibitors, such as sitagliptin (Januvia), saxagliptin (Onglyza), linagliptin (Tradjenta), alogliptin (Nesina), and vildagliptin (Galvus), help increase the proportion of active GLP-1, enhancing insulin secretion. These inhibitors work by competitively binding to DPP-4. This binding causes a significant...
